What it is: P21 (Ac-DGGL(A)G-NH2) is a synthetic, brain-penetrant peptide derived from the active neutralizing region of Ciliary Neurotrophic Factor (CNTF).
How it works: P21 was engineered to bypass CNTF's bulky size and avoid stimulating neutralizing antibodies. It crosses the blood-brain barrier to inhibit leukemia inhibitory factor (LIF) signaling and downregulate anti-neurogenic enzymes. This activates endogenous neurogenesis in the subgranular zone of the dentate gyrus in the hippocampus, stimulating the continuous growth of fresh, healthy neurons and synapses.
