Pharmacological Profile & Mechanism of Action
What it is: PE-22-28 is a synthetic heptapeptide derivative of spadin (a natural propeptide directed against sortilin) specifically engineered as a selective, high-affinity blocker of the two-pore domain potassium channel TREK-1 (K2P2.1).
How it works: By selectively blocking TREK-1 channels in hippocampal neurons, PE-22-28 enhances neuronal excitability, promotes rapid synaptogenesis, and stimulates immediate CREB phosphorylation and BDNF transcription. Unlike conventional monoaminergic modulators, PE-22-28 exerts rapid neurotrophic and behavioral actions within hours rather than weeks.
Why researchers study it: Researched for rapid-onset neuroplasticity, mood elevation, resistance to chronic stress, reversing hippocampal dendritic atrophy, and cognitive preservation in neurodegenerative models.
