Pharmacological Profile & Mechanism of Action
What it is: Pancragen is a synthetic Khavinson tetrapeptide (Lys-Glu-Asp-Trp) specifically targeted at pancreatic islet beta-cells and acinar tissue. It stimulates insulin gene transcription and protects beta-cells from apoptotic exhaustion.
How it works: KEDW binds nuclear chromatin in pancreatic progenitor and islet cells to upregulate pancreatic and duodenal homeobox-1 (PDX-1) transcription factor, enhancing insulin mRNA synthesis. In challenge models, it inhibits caspase-dependent apoptosis and mitochondrial permeability transition pore (mPTP) opening under high-glucose and palmitate-induced lipotoxicity.
