GDF-8 (Growth Differentiation Factor 8 Propeptide) is the endogenous 243-amino-acid prodomain of myostatin that binds non-covalently to mature myostatin, holding it in a latent, inactive conformation. By preventing myostatin from binding to ActRIIB/ALK4/5 receptor complexes, GDF-8 propeptide halts downstream Smad2/Smad3 phosphorylation, releasing skeletal muscle cells from negative growth constraints in cachexia and sarcopenia assays.
