Pharmacological Profile & Mechanism of Action
What it is: Recombinant Somatropin is a 191-amino-acid single-chain polypeptide hormone identical in sequence and tertiary conformation to native human pituitary growth hormone. Synthesized with two critical intramolecular disulfide bridges (Cys53–Cys165 and Cys182–Cys189), this authentic 191AA formulation is free of the non-human N-terminal methionine found in older 192AA somatrem variants, eliminating anti-hGH antibody formation.
How it works: Somatropin binds extracellular Growth Hormone Receptors (GHR), inducing receptor homodimerization and intracellular signaling via the JAK2/STAT5b pathway. In hepatic tissue, this cascades into transcription and secretion of Insulin-like Growth Factor 1 (IGF-1) alongside ternary circulating carriers (IGFBP-3 and ALS). Concurrently, Somatropin exerts direct non-IGF-1-dependent lipolytic action in adipocytes by activating hormone-sensitive lipase (HSL) and downregulating lipoprotein lipase (LPL). In musculoskeletal tissues, it upregulates type I and type III collagen synthesis, stimulates chondrocyte proliferation in articular cartilage, and enhances cellular nitrogen retention.
Why researchers study it: Researched extensively in cellular models of tendon, ligament, and fascial extracellular matrix regeneration, deep slow-wave somatotropic pulsatility, lipid oxidation without glycemic degradation, and endocrine longevity signaling.
